چکیده

While this is available for murine HFs (www.hair-gel.net), systematic characterization of the transcriptome specific human hair follicle (HF) compartments remains a major unmet challenge in research. Here, we have aimed to initiate generation such an atlas reveal HF compartment-specific transcriptome. We performed laser capture microdissection (LCM) on 8 selected compartments, i.e. bulge outer root sheath (ORS), suprabulbar ORS, inner (IRS), precortical matrix (HM), germinative HM, dermal papilla (DP), lower cup (DC), and connective tissue (CTS), anagen VI scalp HFs, analysed these by RNAseq. Principal component analysis (PCA) was used validate distinctness microdissected compartments. Biological replicates were clearly separated high degree similarity distinct transcriptional profiles each examined compartment seen. Afterwards, matched well-known signature genes correct For example, VCAN (versican) ALPL (alkaline phosphatase) correctly expressed DP, DC, CTS, while TCHH (trichohyalin) solely found proximal IRS, KRT85 (Keratin 85) KRT31 31) almost exclusively HM. These transcriptomic data generate easily accessible database identifying defining gene expression profiles, which can be complemented over time ultimately made publicly accessible. This map associated interactive will support identification genes, facilitates development compartment-specific, therapeutic interventions management disorders.

Hair follicle transcriptome profiles during the transition from anagen to catagen in Cashmere goat (Capra hircus).

Previous molecular genetic studies of the goat hair life cycle have focused primarily on a limited number of genes and proteins. To identify additional genes that may play important roles in hair follicle cycle regulation, Illumina sequencing technology was used to catalog differential gene expression profiles in the hair growth cycle (anagen to catagen) of goat, comparing the primary hair foll...

Transcriptome analysis in maritime pine using laser capture microdissection and 454 pyrosequencing.

Maritime pine (Pinus pinaster Aiton) is one of the most advanced conifer models for genomics research. Conifer genomes are extremely large and major advances have recently been made in the characterization of transcriptomes. The combination of laser capture microdissection (LCM) and next-generation sequencing is a powerful tool with which to resolve the entire transcriptome of specific cell typ...

Laser Capture Microdissection in Dentistry

Laser capture microdissection (LCM) allows for the microscopic procurement of specific cell types from tissue sections that can then be used for gene expression analysis. According to the recent development of the LCM technologies and methodologies, the LCM has been used in various kinds of tissue specimens in dental research. For example, the real-time polymerase-chain reaction (PCR) can be pe...

Laser capture microdissection in pathology.

The molecular examination of pathologically altered cells and tissues at the DNA, RNA, and protein level has revolutionised research and diagnostics in pathology. However, the inherent heterogeneity of primary tissues with an admixture of various reactive cell populations can affect the outcome and interpretation of molecular studies. Recently, microdissection of tissue sections and cytological...
